Biochemical changes during in vitro root formation in Chlorophytum borivilianum were investigated in the callus derived from bud pedicel on MS medium supplemented with 4.4 μM BAP + 1.0 mg/l NAA. The callus subcultured on MS medium supplemented with 4.4 μM BAP + 2.69 μM NAA showed roots differentiation within 4–5 days of inoculation. Starch content and reducing sugars were lower while total soluble sugars, free amino-acids, total soluble proteins and total phenols were higher in the roots differentiating callus cultures than in control callus. The activity of enzymes viz. α-amylase, acid-protease, acid-phosphatase and peroxidase increased on 4th and 5th day whereas, the acid invertase activity decreased till the appearance of roots indicating their role in the differentiation.
