Abstract

Agarose and polyacrylamide gel electrophoresis are the two common techniques used to determine the size, purity and sequence identity of DNA fragments. Nucleic acids can also be visualized on gels using fluorescent or silver staining procedures. DNA markers are one of the tools to determine the size of unknown DNA fragments and verify the separation and staining efficiency on gel but are costly. Therefore, a 100 bp DNA ladder was prepared in laboratory by amplifying pUC 19 plasmid template with the help of 2 forward and 5 reverse primers in a cost effective manner.
