In the current work, attempts were made to investigate culture of epicotyls segments with three different cuts (i.e. transverse cut, oblique cut, longitudinal cut), hypocotyls and cotyledon segments derived from in vitro seedlings of pectinifera rootstock. Effect of several factors i.e. culture medium, explants types on regeneration response were examined based on parameters, percentage of explants producing shoots, number of shoots per explants, shoot length and leaf density. Culture of explants on MT medium supplemented with different growth regulators gave rise to disparate regeneration in which MT + BAP (3 mg l-1) was shown to be most effective. Among different explants epicotyls segments was most effective for in vitro shoot regeneration. The transfer of in vitro shoots to rooting media with different concentration of IAA produced complete plantlets where highest rooting percentages were obtained on MS media containing 0.5 mg l-1 IAA. The study can help in developing transgenics of pectinifera rootstocks.
