Doubled haploidy is one of the fundamental tools of plant breeding which accelerates plant breeding and develops homozygous lines in the shortest possible time. About 40,360 anthers from forty one combinations of elite basmati and non-basmati crosses were cultured on five different callus induction media comprising of N6 supplemented with five different combinations of growth regulators. A significant improvement in callus induction was achieved with callus induction media D (dicamba (2.0 mgL−1) + bAP (0.5 mgL−1)) and E (dicamba (2.5 mgL−1) + bAP (0.5 mgL −1)). Addition of dicamba to the media resulted in early and enhanced callus response up to 53.75% but the callus was whitish, non-embryogenic type. Five different plant regeneration media combinations in which MS media was supplemented with different combinations of plant growth regulators were also studied. MSI and MSII plant regeneration media (MS+ kinetin (0.5 mgl−1) + bAP (1.5 mgl−1) + NAA (0.5 mgl−1) and MS +kinetin (0.5 mgl−1) + bAP (1.5 mgl−1) + NAA (0.5 mgl−1) + TDZ (0.5 mgl−1)) were found best for future use.
