Response surface methodology as a statistical tool for process parameter optimization in lipase production by Brevibacterium halotolerans PS4 Isolated from olive orchard soil

Abstract

Astudy was conducted to work out optimization approach forproduction of extracellularlipase from Brevibacterium halotolerans PS4 through response surface methodology, whereas the statistical model was obtained by central composite design. The analysis of variance revealed that the established model was significant (p < 0.01). The influence of various physico-chemical parameters viz. temperature, pH, inoculum size and incubation period on lipase production was examined. The optimum levels of parameters were 40°C temperature, pH 7.0, and 10% inoculum incubated for 74 h for obtaining a maximum lipase activity of 380 IU/ml. The results also demonstrated the feasibility of statistical design tools for optimization of process parameters for maximum lipase production.
