Process optimization of protein isolates derived from chicken heart by pH-shift method and its SDS profiling

Abstract

The aim of the study was to isolate and characterize protein from chicken heart by pH-shift method as no reports are available on the utilization of poultry heart for protein isolates preparation and the manufacture has been impeded by a lack of appropriate technology for their efficient release and process optimization. Initially, the maximum solubility of chicken heart muscle protein was analyzed. Aapproximately 500 g of cleaned from broiler chicken’s heart was used in the experiments. The solubility was found higher at the extreme pH condition i.e., on both side acidic as well as basic and least at isoelectric pH. The highest solubility was recorded at pH of 2 for acidic and 11.5 for alkaline. As compared to alkaline pH (11.5), a significantly higher (p< 0.05) recovery yield of proteins (72.57%) in terms of sediment and lyophilised form. The SDS page analysis also revealed that there is breakdown of native protein to lower molecular weight peptides due to exposure of proteins to extreme pH conditions. Therefore, acidic pH treatment could be an alternative and novel approach for the recovery of protein isolate from chicken heart with higher yield.
