Effect of Cajannus cajan against chlorpyriphos toxicity on isolated chicken hepatocytes

Abstract

The study was conducted to investigate the protective effect of Cajannus cajan against chlorpyriphos toxicity on isolated chicken hepatocytes. The effect under incubation of chlorpyriphos (80 and 160 mg/ml) and aqueous extract of plant (100 and 200 ml) on isolated hepatocytes were observed for 1 hour. The hepatocytes were treated with plant extracts at both doses under simultaneous and pre-treatment mode against chlorpyriphos to study the effect on cell viability and enzyme leakage. Chlorpyriphos at 160 mg/ml significantly increased the supernatant ASAT (20.35%), ALAT (321.11%) and LDH (49.52%) activities and decreased ALP (38.76%) activity along with cell viability (26.50%). Pre-treatment of plant extract at 200 ml significantly decreased/increased these enzymes and brought back the cell viability to almost normal in comparison to 100 ml/ml dose and simultaneous treatment mode. Chlorpyriphos, a neurotoxic organophosphate, induced significant hepatocyte damage at 160 mg/ml against which the plant extract at 200 ml/ml dose and under pre-treatment regimen exhibited better protective effect than the counter parts.
