A total of 349 quarters of 89 lactating cows comprising 31 Triple cross (TP) [Kankrej × Jersey × Holstein Friesian (HF)], 29 Kankrej, 17 Gir and 12 HF were screened for subclinical mastitis. A percentage of 52.8 (47/89) of cows and 30.66% (107/349) quarters were found to be positive for subclinical mastitis. A total of 68 Staphylococci could be identified from the 107 positive quarters. Out of 68 Staphylococci isolates, 55.89%, 45.58% and 70.58% isolates showed fermentation on mannitol salt agar, positive for pigment production and positive for coagulase reaction, respectively. Majority (45.58%) isolates exhibited haemolysin production. Maximum numbers of the Staphylococci isolates were lysed by group II and phage number 47. All the Staphylococci isolates were sensitive to methicillin and oxacillin antibiotics. Of the 68 Staphylococci isolates, 30 isolates could be identified as Staphylococcus aureus as they yielded an expected amplification product of 1, 318 bp, while 38 Staphylococci were negative by polymerase chain reaction (PCR).
