Multiplex PCR for virulence associated genes in E.coli isolates from calf diarrhoea

Abstract

A multiplex PCR was designed for the detection of following virulence genes viz. enteropathogenic (eaeA/bfp), enteroinvasive (invE) and enteroaggregative (EAST1) from different categories of diarrheagenic Escherichia coli in calf diarrhea cases. A total of 91 E.coli isolates selected for study belong to certain category of diarrheagenic E.coli viz. eae (intimin) for EPEC, bfp (bundle forming pilli) belong to typical EPEC, EASTI (Enteroaggregative heat stable toxin I) for EAEC, and invE (invasive E.coli) for EIEC. Multiplex PCR based detection of virulence associated genes among 91 E.coli isolates showed the presence of amplified product size of 815bp in 15 (16.8%) isolates indicating the presence of intimin (eae) gene. Eighteen (19.78%) isolates possessed bfp gene of 597bp and 62 (68.13%) isolates harbored EAST1 gene of 111bp and in 15 (16.48%) isolates amplification of 382bp product size indicated the presence of invE gene. The multiplex PCR was found to be specific and useful assay.
