Abstract

The cDNA coding for Granulocyte macrophage colony stimulating factor (GM-CSF) of the Hallikar breed of Indian origin cattle was amplified by reverse transcriptase polymerase chain reaction (RT-PCR) from LPS stimulated macrophages and it was cloned into pCNDA vector and sequenced. The amplified bovine GM-CSF cDNA was 755 bp in length without the secretory signal peptide sequence. The GM-CSF nucleotide sequence was 99 and 96% homologous to Bos taurus and Bubalus bubalis respectively and the 755 bp open reading frame (ORF) coded for 143 amino acids of molecular weight 16200 Da. Phylogenetic analysis showed that GM-CSF sequence of swine and equine are most divergent than buffalo, sheep, goat and deer.
