Caspase Activating Recruitment Domain-15 (CARD-15) gene plays an important role in initiation of inflammatory reactions and subsequent immune response. They have been identified as susceptibility loci for inflammatory bowel disease in humans and are, therefore, suitable candidate gene for inflammatory disease resistance in cattle. As 1988 bp long exon 12 was found to be highly polymorphic in Bos taurus, this study was conducted to explore polymorphism in exon 12 of CARO-15 gene in Bos indicus. Genomic DNA were extracted from 162 lactating Sahiwal cattle maintained at National Dairy Research Institute (NDRI), Kamal (Haryana) and population was screened using PCR-RFLP with Pvull and Sad restriction enzymes. Five sets of primers were used to amplify five overlapping contigs of exon 12 Le. 12.1 to 12.5 (578, 619, 641, 509 and 588 bp). Pvull RE exhibited AA (1.9%), AB (91.9%) and BB (6.2%) genotypes in contig 12.5, whereas, Sad RE revealed only two, AA (46.9%) and AB (53.1%) polymorphic patterns in contig 12.2. Seven SNPs [240 (T>C), 820 (C>T), 1487 (A>G), 1641 (A>G), 1682 (G>A), 1685 (T>A) and 1982 (C>T)] were found in the complete sequence of exon 12 of CARD-15 gene of Sahiwal cattle. It is concluded that there are several SNPs in 3′ UTR of Bos indicus as compared to that of Bos taurus.
